Madekwe was born South London, England into an ethnically diverse family. Madekwe is English but her father is Nigerian. She was born and raised on a council estate south of London. At the age fourteen, she was enrolled at The Brit School. In London, she studied at the Royal Academy of Dramatic Art in which her acting diploma was obtained. She appeared on stage in several productions as a student such as the role of King Henry V, as Princess Katherine, and Wuthering Heights, as Catherine Earnshaw. Madekwe started her acting career through the movie Storm Damage. Over the past few years Madekwe's appearance in Teachers and Doctors. Madekwe landed her first motion-picture role in the Woody Allen film Cassandra's Dream, opposite Colin Farrell and Ewan McGregor. In the following year, she played Elisha (a BBC Three drama-pilot) in West 10 LDN. Additionally, she starred in Trexx as Ollie and in six episodes of Flipside.
